Biệt hóa tế bào chức năng gan từ tế bào gốc người và chuột
Ghép tế bào gốc (TBG) đang trở thành một trong những phương pháp điều trị bệnh đầy tiềm năng cho các bệnh lý thoái hóa, tổn thương hệ miễn dịch hay các tổn thương mất mô, cơ quan. Trong đó, các bệnh liên quan tới gan có thể ghép TBG biệt hóa thành tế bào gan là phổ biến.

Vì vậy, năm 2019, nhóm nghiên cứu của TS Nguyễn Văn Hạnh (Viện Công nghệ Sinh học, thuộc Viện Hàn lâm KH&CN Việt Nam) đã nghiên cứu đánh giá khả năng biệt hóa tạo tế bào chức năng gan và khu trú của TBG trong mô hình cấy ghép TBG điều trị bệnh gan.

Đề tài, đã được nghiệm thu đạt loại xuất sắc, thu được nguồn TBG đầy đủ tiềm năng đặc trưng, nhân nuôi và duy trì được trong điều kiện in vitro, đủ điều kiện cho các nghiên cứu cơ bản và nghiên cứu ứng dụng khác trong tương lai. Đề tài đã tạo được tế bào với các chỉ thị đặc trưng chức năng tế bào gan từ TBG trung mô người và TBG tủy xương chuột khi tiến hành xử lý TBG với các công thức gây biệt hóa khác nhau.

Đề tài cũng đã tạo được mô hình đánh giá khả năng định vị sau cấy ghép TBG, tế bào chức năng gan biệt hóa từ TBG trên mô hình bệnh gan bằng phương pháp gắn TBG với hạt nano kim cương phát quang.

Nghiên cứu này đầu tiên trình bày thông tin liên quan đến cấy ghép TBG, tế bào chức năng gan biệt hóa đã được gắn đánh dấu bằng hạt nano kim cương trên chuột mô hình bệnh gan. Nghiên cứu cũng lần đầu tiên cho thấy hiệu quả của cấy ghép tế bào chức năng gan biệt hóa trong mô hình chuột bệnh gan.
